目的 观察高压氧 (Hyperbaric Oxygen,HBO)对老龄大鼠脑缺血 /再灌注 (Cerebral Ischemia/Reperfusion,CI/ R)后海马神经元超微结构的影响。方法 建立老龄大鼠不完全性脑缺血动物模型。不进行 HBO处置正常对照组 (A组 )、脑缺血 3 0 min再灌注 6h(B组 )、12 h(C组 )、2 4h(D组 )、48h(E组 )、12 d(F组 )和 2 2 d(G组 )。HBO处置组 :将大鼠按不同的再灌注时程暴露于 0 .2 5MPa(2 .5ATA)高压氧环境中 ,每天 1次 ,每次给氧 3 0 min×2 (间歇 10 m in) ,连续 10次 (1疗程 )和 2 0次 (2疗程 )。 1疗程组 :老龄正常对照组 (A1组 )、缺血 3 0 min再灌注 6h (B1组 )、12 h (C1组 )、2 4h (D1组 )、48h(E1组 ) ;2疗程组 :老龄正常对照组 (A2 组 )、缺血 3 0 min再灌注 6h (B2 组 )、12 h(C2组 )、2 4h (D2 组 )、48h(E2 组 )。用透射电镜观察海马神经元的超微结构 ,通过计算机图像分析系统对线粒体形态计量分析。结果 A组、A1组和 A2 组海马的超微结构基本正常 ;CI/ R 6h组 1疗程和 2疗程超微结构改变不明显 ;CI/ R2 4h组 1疗程线粒体肿胀、嵴模糊 ,部分粗面内质网扩张、脱颗粒 ,2疗程后基本恢复正常 ;CI/ R48h组 1疗程体积缩小 ,细胞核内异染色质增多、边聚 ,核周间隙增宽 ,线粒体肿胀、密度降低、嵴模糊 ,?
Objective To observe the effect of Hyperbaric Oxygen (HBO) on the ultrastructure of hippocampal neurons in the aged rats after cerebral ischemia / reperfusion (CI / R). Methods The animal model of incomplete cerebral ischemia in aged rats was established. The rats in control group (group A), reperfusion group 6h (group B), 12 h (group C), 24 h (group D), 48 h (group E), 12 d Group) and 22 days (group G). HBO treatment group: The rats were exposed to 0,25 MPa (2.5 ATA) hyperbaric oxygen atmosphere at different reperfusion durations once a day for 30 min × 2 (intermittent 10 min) 10 times in a row (1 course) and 20 times (2 courses). 1 course of treatment: aged normal control group (A1), ischemia reperfusion 6 min (B1), 12 h (C1), 24 h (D1), 48 h The aged normal control group (A2), ischemia reperfusion 6 min (B2), 12 h (C2), 24 h (D2), 48 h (E2). The ultrastructure of hippocampal neurons was observed by transmission electron microscope, and the morphology of mitochondria was analyzed by computer image analysis system. Results The ultrastructure of hippocampus in group A, group A1 and group A2 were basically normal. There was no obvious change in ultrastructure in the course of 1 course and 2 courses in CI / R 6h group. The mitochondrial swelling, The endoplasmic reticulum was dilated and degranulated and returned to normal after 2 courses of treatment.
