目的:通过恒化器生物膜模型比较五倍子和洗必泰的抗菌效果,为中药替代化学药物预防龋病的应用推广提供理论依据。方法:建立恒化器牙菌斑生物膜模型,经五倍子和洗必泰处理后,通过生物膜上细菌计数、连续pH的监测、生物膜超微结构观察,比较二者对口腔常见细菌的生长、产酸和生物膜表面形态的影响。结果:①五倍子和洗必泰对实验菌株的生长都有不同程度的抑制,五倍子对生物膜细菌的生态组成无明显影响。②二者都能够抑制实验细菌产酸,使恒流培养室内的终末pH高于临界pH;4 g.L-1五倍子抑制产酸的能力同0.5 g.L-1洗必泰相近。③与对照组相比,五倍子和洗必泰处理后的生物膜表面菌细胞界限清晰、绒团样胞外基质相对减少,二者间未见明显差别。结论:在生物膜模型中,五倍子可以通过抑制细菌生长、产酸、产胞外基质等途径,抑制生物膜细菌的生长代谢,4 g.L-1五倍子的抗菌效果同0.5 g.L-1洗必泰无明显差异。提示五倍子是一种具有应用前景的防龋中药。
Objective: To compare the antibacterial effects of gallnut and chlorhexidine by chemostat biofilm model, and provide theoretical basis for the application and promotion of alternative medicine for traditional Chinese medicine to prevent rickets. METHODS: A chemostat dental plaque biofilm model was established. After treatment with Galla Chinensis and chlorhexidine, the bacteria were counted on a biofilm, continuous pH was monitored, and the ultrastructure of the biofilm was observed to compare the growth of oral common bacteria. , acid production and biofilm surface morphology effects. Results: 1 Chinese gallnut and chlorhexidine had different degrees of inhibition on the growth of experimental strains. Both can inhibit the acid production of experimental bacteria, so that the terminal pH in the constant current culture chamber is higher than the critical pH; the ability of 4 g.L-1 gallons to inhibit acid production is similar to that of 0.5 g.L-1 chlorhexidine. 3 Compared with the control group, gallbladder and chlorhexidine-treated biofilms had a clear boundary of the surface bacteria cells and a relatively decreased tuft-like extracellular matrix, and there was no significant difference between the two. Conclusion: In the biomembrane model, Galla chinensis can inhibit the growth and metabolism of biofilm bacteria by inhibiting bacterial growth, acid production, extracellular matrix production, etc. The antibacterial effect of 4 gL-1 gallnut with 0.5 gL-1 chlorhexidine Significant differences.
