目的:优选超声提取鸡冠花总黄酮的提取工艺,并测定其不同产地、采收期总黄酮含量。方法:以总黄酮含量为考察指标,采用单因素试验和正交试验优选鸡冠花中总黄酮的提取工艺,所得结果进行方差分析。采用优选最佳超声提取工艺比较测定10个不同产地及8-12月中旬采收的鸡冠花中总黄酮含量。结果:优选出超声辅助提取鸡冠花总黄酮的最佳工艺条件为甲醇用量50mL,提取温度为40℃,提取时间为60min;10个不同产地鸡冠花含量差异较大,安徽、河北所产鸡冠花总黄酮含量较高;11月采收的鸡冠花总黄酮含量较高。结论:该测定方法简便、快捷,其测定结果可为鸡冠花的质量控制及合理应用提供一定的依据。
OBJECTIVE: To optimize the extraction technology of total flavonoids from Cercaria, determine the content of total flavonoids in different producing areas and harvesting period. Methods: Taking total flavonoids content as index, single factor experiment and orthogonal experiment were used to optimize the extraction technology of total flavonoids in celosia. The results were analyzed by ANOVA. The best optimized ultrasonic extraction process was used to determine the content of total flavonoids in celosia collected from 10 different producing areas and from the middle of August to the middle of December. Results: The optimum extraction conditions of ultrasonic-assisted extraction of total flavonoids from Cercis chinensis were 50mL methanol, 40 ℃ extraction temperature and 60min extraction time. The content of celosia in 10 different producing areas varied greatly. The content of total flavonoids is higher; the content of total flavonoids in celosia collected in November is higher. Conclusion: The method is simple and rapid, and the determination results can provide a basis for the quality control and rational application of Celosia.
