目的 :用缬沙坦 ( valsartan)和苯那普利 ( benazepril)干预自发性高血压大鼠 ( SHR) ,观察大鼠主要组织内肾素—血管紧张素活性变化。 方法 :将 SHR随机分成 4组 (生理盐水组、苯那普利组、小剂量缬沙坦组、大剂量缬沙坦组 ,各组 n=6) ,并设 WistarKyoto( WKY)大鼠为血压正常对照组 ( WKY大鼠对照组 ,n=6) ,用放射免疫测定法观察各组血浆和主要器官组织内肾素—血管紧张素活性和血管紧张素 ( Ang )水平。 结果 :1SHR的 4组心、脑、肾、肝中肾素活性较 WKY大鼠对照组均明显增高 ( P<0 .0 5 ) ,组织中 Ang 水平亦较WKY大鼠对照组增高。 2用苯那普利组干预 4周后的 SHR,血浆和肾组织中肾素活性较生理盐水组明显升高 ,而血浆和各组织中 Ang 水平均有轻度下降 ( P>0 .0 5 )。3大剂量缬沙坦组、小剂量缬沙坦组干预 4周后的 SHR血浆和心、肾、肝组织中肾素活性较生理盐水组增高 ( P<0 .0 5或 <0 .0 1) ,且 SHR心、肾、肝组织中肾素活性大剂量缬沙坦组干预 4周后又明显高于苯那普利组 ;大剂量缬沙坦组、小剂量缬沙坦组血浆、各组织 Ang 水平均明显增高 ( P<0 .0 5或 <0 .0 1)。 结论 :实验表明 SHR血浆和组织内肾素—血管紧张素系统较 WKY大鼠活跃。用血管紧张素转换酶抑制剂和 Ang 的 型受体拮抗剂
OBJECTIVE: To investigate the changes of renin-angiotensin receptor (ACE) activity in major tissues of spontaneously hypertensive rats (SHR) with valsartan and benazepril. Methods: SHR were randomly divided into 4 groups (normal saline group, benazepril group, low dose valsartan group, high dose valsartan group, each group n = 6), and WistarKyoto (WKY) The normal control group (WKY rat control group, n = 6), the radioimmunoassay was used to observe the renin-angiotensin (ACE) and angiotensin (Ang) levels in plasma and major organs of each group. Results: The activities of renin in heart, brain, kidney and liver of 1SHR group were significantly higher than those of WKY control group (P <0.05), and the level of Ang in the tissue was also higher than that of WKY rat control group. 2 After 4 weeks intervention with benazepril, renin activity in plasma and renal tissue was significantly increased compared with that in saline group, while the levels of Ang in plasma and each tissue were slightly decreased (P> 0.05) ). Renal activity of SHR plasma and heart, kidney and liver tissue of high-dose valsartan group and low-dose valsartan group were higher than those of saline group after 4 weeks of intervention (P <0.05 or <0.01 ), And high-dose renin-active valsartan group in SHR hearts, kidneys and liver tissues was significantly higher than the benazepril group after 4 weeks of intervention; high-dose valsartan group, low-dose valsartan plasma Tissue Ang levels were significantly higher (P <0.05 or <0.01).
