目的:通过实验观察道地通管汤对输卵管炎性大鼠信号转导通路TGF-β1、Smad2、Smad3、Smad7蛋白的影响,探讨其作用机制。方法:80只SD雌性大鼠随机抽取12只做正常组,剩余大鼠采用混合菌制成输卵管炎性模型后将其分为模型组、康妇消炎栓组及道地通管汤低、中、高剂量组。每组采用相应的干预措施灌肠,正常组和模型组予生理盐水0.2 m L/100 g·d,阳性对照组予康妇消炎栓水溶剂0.2 m L/100 g·d,道地通管汤低、中、高剂量组分别予0.35 g/100 mg·d、0.7 g/100 mg·d、1.4 g/100 g·d。于灌肠第15天、30天采集输卵管,蛋白印迹法(Western blotting)检测输卵管组织TGF-β1、Smad2、Smad3及smd7蛋白的表达。结果:1与正常组比较,模型组及各治疗组TGF-β1、Smad2、Smad3的表达量增加,Smad7的表达量减少(P<0.05);2与模型组比较,各治疗组能不同程度降低模型大鼠输卵管组织中TGF-β1、Smad2、Smad3表达量,增加Smad7的表达量,以道地通管汤高剂量最为突出(P<0.05);3各组自身比较,模型组30 d时TGF-β1、Smad2、Smad3的表达量高于15 d的,而Smad7的表达量低于15 d(P<0.05);各治疗组给药30 d时TGF-β1、Smad2、Smad3的表达量低于给药15 d的,而Smad7的表达量高于给药15 d,以道地通管汤高剂量最为突出(P<0.05)。结论:道地通管汤对输卵管炎性大鼠有明显的治疗作用,其机制可能通过TGF-β1/Smads信号通路下调输卵管组织中TGF-β1、Smad2、Smad3表达,上调Smad7的表达,抑制炎症反应、减轻输卵管组织纤维化,从而改善输卵管组织结构。
OBJECTIVE: To observe the effect of daotiedongzhi decoction on TGF-β1, Smad2, Smad3 and Smad7 protein in tubal inflammatory signal transduction pathway in rats and to explore its mechanism. The expression of TGF-β1, Smad2 and Smad3 in each treatment group was lower than Administration of 15 d, while the expression of Smad7 was higher than the administration of 15 d, the highest dose of Dantu Tong Guan Tang the most prominent (P <0.05). Conclusion: Daodi Tongzhi Decoction has obvious curative effect on tubal inflammatory rats, and its mechanism may down-regulate the expression of TGF-β1, Smad2 and Smad3 in tubal tissue by TGF-β1 / Smads signal pathway, up-regulate the expression of Smad7 and inhibit the inflammation Reaction, reduce tubal tissue fibrosis, thereby improving the tubal tissue structure.
