目的 建立和测试低温冷冻血小板无电源运输保存技术。方法 采用高效发泡材料为主要隔热材料 ,以食品级干冰砖为冷源 ,低温冷冻血小板与干冰砖体积比为 2 :1,共同置于箱内保存。记录温度变化曲线和箱温分别在 -70℃ ,-4 0℃ ,-2 0℃以下的维持时间 ;60袋 -80℃冰箱保存低温冷冻血小板按上述方法保存至 -4 0℃后重新放回 -80℃冰箱保存。随机取始终保存于 -80℃冰箱冷冻血小板为对照组 ,取测试冷冻血小板其中 2 0袋为测试组 ,比较两组血小板计数、血小板平均体积 (MPV)、血小板分布宽度 (PDW )、血浆乳酸脱氢酶 (LDH)、乳酸、磷脂酰丝氨酸 (PS)阳性率和CD62p再表达率。结果 在 2 3℃~ 2 6℃环境无电条件下将低温冷冻血小板的保存温度在 -70℃以下可维持 95h ;-4 0℃以下可维持 113h ;-2 0℃以下可维持 118h。测试组和对照组血小板计数、MPV、PDW、LDH、乳酸、pH、PS阳性率和CD62p再表达率差异无显著性。结论 无电源条件下低温冷冻血小板和新鲜冰冻血浆储存运输技术 ,满足在低温冷冻血小板和新鲜冰冻血浆在野战医院的应用所需的基本条件。
Objective To establish and test cryogenic frozen platelets without power transport and storage technology. Methods High-performance foamed material was used as the main insulation material. The food-grade dry ice brick was used as the cold source. The volume ratio of cold-frozen platelets to dry-ice brick was 2: 1. Record the temperature curve and the box temperature at -70 ℃, -4 0 ℃, -2 0 ℃ the following holding time; 60 bags -80 ℃ refrigerator preservation of frozen frozen platelets stored in accordance with the above method to -4 ℃ and then back -80 ℃ refrigerator to save. Randomly stored at -80 ℃ frozen frozen platelets as a control group, frozen frozen platelets to test 20 bags as the test group, comparing the platelet count, platelet mean volume (MPV), platelet distribution width (PDW), plasma lactate Hydrogenase (LDH), lactate, phosphatidylserine (PS) positive rate and CD62p re-expression rate. Results The preservation temperature of cryopreserved platelets was maintained at -70 ℃ for 95h under the condition of no electricity from 23 ℃ ~ 26 ℃. The temperature maintained below -4 ℃ for 113h and the duration below -2 ℃ for 118h. There was no significant difference of platelet count, MPV, PDW, LDH, lactate, pH, PS positive rate and CD62p re-expression rate between test group and control group. Conclusion Low temperature frozen platelets and fresh frozen plasma storage and transportation technology without power supply can meet the basic requirements for the application of cryogenic frozen platelets and fresh frozen plasma in field hospitals.
