目的:研究四物合剂逆转人红白血病细胞株K562/ADM多药耐药作用的药效活性组分。方法:用大孔吸附树脂分离四物合剂,并用体外细胞模型作药效评价,寻找发现活性部位,再用HPLCMS方法研究活性部位的化学组成,进而确定其主要活性组分。结果:熟地黄粗多糖对K562/ADM的耐药性有明显的逆转作用,其逆转倍数与对照组(无逆转剂)相比有显著差异(P<0.01),并且与主要活性部位的逆转作用无明显差异(P>0.05)。结论:熟地黄粗多糖能逆转K562/ADM细胞的多药耐药性,是四物合剂逆转K562/ADM多药耐药性的主要活性组分之一。
Objective: To study the efficacy of Siwu Mixture in reversing the multidrug resistance of human leukemia cell line K562/ADM. METHODS: Macroporous adsorption resin was used to separate Siwu Mixture. The in vitro cell model was used for evaluation of drug efficacy. The active site was found and then the HPLCMS method was used to study the chemical composition of the active site. The main active components were then determined. RESULTS: The crude polysaccharide of Rehmannia glutinosa had a significant reversal effect on the drug resistance of K562/ADM. The reversal multiple was significantly different from that of the control group (no reversal agent) (P<0.01), and was reversed from the main active site. No significant difference (P>0.05). CONCLUSION: Rehmannia glutinosa polysaccharide can reverse the multidrug resistance of K562/ADM cells, and it is one of the main active components of Siwu Mixture in reversing the multidrug resistance of K562/ADM.
