目的 观察胶质细胞源性神经营养因子 (GDNF)及单纯疱疹病毒 (HSV)载体介导的 GDNF(HSV-GDNF)对坐骨神经损伤大鼠脊髓运动神经元 Bcl- 2表达的影响。 方法 分别取坐骨神经损伤后 4d、7d和 14d大鼠(分成对照组、GDNF组和 HSV- GDNF组 )的腰段脊髓 (L4~ 6 ) ,行石蜡包埋、切片 ;用抗 Bcl- 2抗血清进行免疫组织化学染色 ,观察 Bcl- 2免疫反应 (Bcl- 2 - IR)神经元数目 ,并在图像分析仪上对 Bcl- 2 - IR阳性神经元作光密度的色谱分析。 结果 1.坐骨神经损伤后 4d、7d时 ,GDNF组和 HSV- GDNF组损伤侧脊髓运动神经元 Bcl- 2 - IR阳性神经元的数量和平均光密度均明显高于对照组损伤侧。2 .坐骨神经损伤后 14d时 ,对照组、GDNF组和 HSV- GDNF组损伤侧脊髓运动神经元对 Bcl- 2的表达已无明显差别。 结论 GDNF与 HSV- GDNF能够增强坐骨神经损伤大鼠脊髓运动神经元 Bcl- 2的表达 ,减少神经元的退化死亡。
Objective To investigate the effect of GDNF (HSV-GDNF) mediated by glial cell line-derived neurotrophic factor (GDNF) and herpes simplex virus (HSV) on the expression of Bcl-2 in spinal cord motoneurons of rats with sciatic nerve injury. Methods The lumbar spinal cord (L4 ~ 6) of rats in 4d, 7d and 14d after sciatic nerve injury (divided into control group, GDNF group and HSV-GDNF group) were respectively paraffin-embedded and sectioned. Immunohistochemical staining was performed to observe the number of Bcl - 2 - immunoreactive (Bcl - 2 - IR) neurons. The optical density of Bcl - 2 - IR positive neurons was analyzed by an image analyzer. The number and average optical density of Bcl-2 - IR positive neurons in the injured spinal cord of GDNF group and HSV-GDNF group were significantly higher than those of the control group 4 and 7 days after sciatic nerve injury. On the 14th day after sciatic nerve injury, the expression of Bcl-2 in injured spinal cord motor neurons of control group, GDNF group and HSV-GDNF group had no significant difference. Conclusion GDNF and HSV-GDNF can enhance the expression of Bcl-2 in spinal motor neurons of sciatic nerve injury rats and reduce the degeneration of neurons.
