目的探讨阿司匹林对糖尿病大鼠外周血T淋巴细胞及相关细胞因子和免疫器官系数的影响。方法链脲佐菌素腹腔内注射雄性SD大鼠诱导糖尿病,分为糖尿病组(DM组n=12)和糖尿病阿司匹林干预组(AD组n=12),另有鼠龄和体重相似的雄性SD大鼠作为对照组(NC组n=12)。AD组用阿司匹林30mg﹒kg-1﹒d-1灌胃,DM组和NC组用等量的蒸馏水灌胃;流式细胞术检测CD3+T细胞、CD4+T细胞以及CD8+T细胞比例,酶联免疫吸附法(ELISA)测定各组大鼠血浆中TNF-α及IL-6,无菌取胸腺和脾脏计算免疫器官系数。结果实验12周时,DM组CD3+T细胞、CD4+T细胞、CD8+T细胞和胸腺指数,显著低于NC组(P<0.01),血浆中IL-6和TNF-α显著高于NC组(P<0.01)。与DM组相比,AD组CD8+T细胞无显著改变(P>0.05),但AD组CD3+T细胞、CD4+T细胞和胸腺指数显著高于DM组(P<0.01),血浆中TNF-α和和IL-6显著低于DM组(P<0.01)。结论阿司匹林对糖尿病大鼠免疫系统有保护作用。
Objective To investigate the effect of aspirin on peripheral blood T lymphocytes, related cytokines and immune organ in diabetic rats. Methods Streptozotocin (STZ) was injected intraperitoneally into male Sprague-Dawley rats to induce diabetes mellitus. The diabetic SD rats were divided into diabetic group (n = 12 in DM group) and diabetic aspirin intervention group (n = 12 in AD group) Rats served as control group (NC group n = 12). AD group with aspirin 30mg. kg-1. The mice in DM group and NC group were given gavage with equal volume of distilled water. The proportion of CD3 + T cells, CD4 + T cells and CD8 + T cells were measured by flow cytometry. The percentage of CD8 + T cells in each group was determined by enzyme-linked immunosorbent assay (ELISA) The levels of TNF-α and IL-6 in the plasma of the rats were measured, and the immune organ coefficients were calculated from the thymus and spleen of the rats. Results At 12 weeks, the levels of CD3 + T cells, CD4 + T cells, CD8 + T cells and thymus in DM group were significantly lower than those in NC group (P <0.01), and the levels of IL-6 and TNF- Group (P <0.01). Compared with DM group, CD8 + T cells in AD group had no significant change (P> 0.05), but CD3 + T cells, CD4 + T cells and thymus index in AD group were significantly higher than those in DM group (P <0.01) -α and IL-6 were significantly lower than those in DM group (P <0.01). Conclusion Aspirin has a protective effect on the immune system in diabetic rats.
