目的:通过观察针药结合对心肌缺血再灌注大鼠模型血清中CK、LDH含量变化的影响,以及对心肌组织中IL-6、IL-8及TNF-α含量变化的影响,揭示针药结合干预心肌缺血再灌注损伤的作用机制,为临床选用更有效的干预措施提供佐证。方法:采用随机数字表法将60只健康雄性SD大鼠随机分成空白对照组、模型对照组、中药复方组、针药结合组、西药对照组,每组12只。除空白对照组外,其他各组均冠脉结扎法建立心肌缺血再灌注大鼠模型。造模成功后,给予相应的干预措施,每天1次,1周后应用酶联免疫吸附法(ELISA)检测血清中CK、LDH的表达水平,并且检测心肌组织中IL-6、IL-8及TNF-α的表达水平。结果:与空白对照组比较,模型对照组大鼠血清中CK、LDH的含量和心肌组织中IL-6、IL-8及TNF-α的含量都显著升高(P<0.05)。其它各组经相应的干预措施治疗后,各组大鼠血清中CK、LDH的含量和心肌组织中IL-6、IL-8及TNF-α的含量都显著降低,与模型对照组有显著性差异(P<0.05),其中,针药结合组大鼠的效果最佳,与各组比较也存在显著性差异(P<0.05)。结论:中药复方、针药结合及西药均可有效降低心肌缺血再灌注大鼠血清中CK、LDH的含量和心肌组织中IL-6、IL-8及TNF-α的含量,但以针药结合组效果最佳。
OBJECTIVE: To observe the effects of acupuncture and medicine on the changes of CK and LDH contents in myocardium of myocardial ischemia-reperfusion rats and the changes of IL-6, IL-8 and TNF-α contents in myocardium, Combined with the mechanism of intervention in myocardial ischemia-reperfusion injury, to provide evidence for clinical use of more effective interventions. Methods: Sixty healthy male Sprague-Dawley rats were randomly divided into blank control group, model control group, TCM compound group, acupuncture combination group and western medicine control group, with 12 rats in each group. Except the blank control group, all the other groups were established myocardial ischemia-reperfusion rat model by coronary ligation. After successful modeling, the corresponding interventions were given. The levels of CK and LDH in serum were detected by enzyme linked immunosorbent assay (ELISA) after 1 and 1 week respectively. The levels of IL-6, IL-8 and TNF-α expression levels. Results: Compared with the blank control group, the levels of serum CK and LDH and the levels of IL-6, IL-8 and TNF-α in myocardial tissue of model control group were significantly increased (P <0.05). After the other groups were treated by the corresponding interventions, the content of CK, LDH and the content of IL-6, IL-8 and TNF-α in the serum of the rats in each group were significantly decreased, which were significantly compared with the model control group (P <0.05). Among them, the acupuncture-drug combination group had the best effect and there was significant difference compared with each group (P <0.05). Conclusion: The Chinese medicine compound, acupuncture combined with medicine and western medicine can effectively reduce the content of CK, LDH and the contents of IL-6, IL-8 and TNF-α in myocardium of myocardial ischemia-reperfusion rats, The best combination effect.
