目的探讨脑室内低温对家兔大脑中动脉梗塞模型脑组织肿瘤坏死因子-α(TNF-α)与白介素-1β(IL-1β)表达的影响。方法选用4~6个月龄雄性家兔20只,随机分为对照组(37℃常温生理盐水)和低温组(22℃低温生理盐水),各10只。应用线栓法建立大脑中动脉闭塞(MCAO)模型,进行脑室内穿刺低温液体灌注亚低温干预,24 h后,参照Zea Longa及Bederson检查法进行评分。免疫组织化学方法和Western blot方法检测各组脑组织TNF-α与IL-1β的表达。结果兔神经病学体征评分以及动物行为观察发现,低温组评分明显优于常温组(P<0.01)。与对照组相比,低温组家兔脑组织的TNF-α与IL-1β的表达显著降低(P<0.01)。结论脑室内穿刺低温干预可能通过调节TNF-α与IL-1β的表达对急性脑梗塞家兔动物模型提供脑保护作用。
Objective To investigate the effect of intraventricular hypothermia on the expression of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in rabbits with middle cerebral artery occlusion (MCAO) model. Methods Twenty male rabbits of 4 to 6 months old were randomly divided into control group (37 ℃ normal saline) and low temperature group (22 ℃ hypothermia saline), each 10 rabbits. The middle cerebral artery occlusion (MCAO) model was established by the method of thread occlusion, and the intra-ventricle puncture hypothermia was used to perfusion hypothermia intervention. After 24 h, the patients were scored according to the Zea Longa and Bederson test. Immunohistochemistry and Western blot were used to detect the expression of TNF-α and IL-1β in each group. Results The score of neurological examination in rabbits and the observation of animal behavior showed that the score of hypothermia group was significantly better than that of normal temperature group (P <0.01). Compared with the control group, the expression of TNF-α and IL-1β in the brain tissue of hypothermia group was significantly decreased (P <0.01). Conclusion Low-temperature intra-ventricular puncture may provide cerebral protection in animal models of acute cerebral infarction by regulating the expression of TNF-α and IL-1β.
