目的:分析刺五加叶片、叶柄及其愈伤组织中SS、SE和bAS基因表达量的差异。方法:以刺五加叶片和叶柄为外植体,诱导获得愈伤组织。实时荧光定量PCR检测SS、SE和bAS基因的表达量。结果:刺五加SS、SE和bAS基因在刺五加叶片、叶柄及其愈伤组织中都有表达,SS、SE和bAS基因在叶片愈伤组织中的表达量分别为叶片中的89.3%、73.8%和83.4%,在叶柄愈伤组织中的表达量分别为叶柄中表达量的80.2%、87.7%和70.9%。结论:刺五加愈伤组织中SS、SE和bAS基因的表达量显著低于植株中的表达量。
Objective: To analyze the difference of SS, SE and bAS gene expression in leaves, petioles and callus of Acanthopanax senticosus. Methods: The leaves and petiole of Acanthopanax senticosus were used as explants to induce callus. Real-time fluorescence quantitative PCR was used to detect the expression of SS, SE and bAS genes. Results: The SS, SE and bAS genes of Acanthopanax senticosus were expressed in leaves, petioles and callus of Acanthopanax senticosus. The expression levels of SS, SE and bAS in leaf callus were 89.3% , 73.8% and 83.4% respectively. The expression levels in petiole callus were 80.2%, 87.7% and 70.9% respectively. Conclusion: The expression levels of SS, SE and bAS in calli of Acanthopanax senticosus were significantly lower than those in plants.
