随着科研项目的广泛开展,要显示组织内的特殊结构,如脂肪组织或类脂等,需要采用冷冻切片。通常冷冻切片采用液体二氧化碳和氯乙烷等作制冷剂,目前也采用半导体致冷调节器来代替液体二氧化碳的冰冻。这些方法都要有一套特有的设备,并且操作起来也较繁复。我们采用液氮作制冷剂,对小鼠肝脏行冰冻切片,做脂肪染色,效果满意。做法: 1.取经给药后处死的小鼠肝脏,固定手中性甲醛液(内含90%生理盐水和10%福尔马林)中,2小时后将肝脏切开,以便充分固定,24小时后可行冰冻切片。 2.将一块湿滤纸平铺予标本头子(一般切片机均
