目的:观察氯沙坦对肝星状细胞凋亡的影响。方法:双染法流式细胞仪(AnnexinV-FITC/PI)检测肝星状细胞的凋亡率;免疫组化法检测Bcl-2、Bax。结果:双染法检测显示,各浓度的氯沙坦均能促进细胞凋亡,与对照组比较差异有统计学意义(P<0.01)。免疫组化检测显示氯沙坦可引起Bcl-2表达下降,Bax表达上升,与对照组比较差异有统计学意义(P<0.05)。结论:氯沙坦有诱导肝星状细胞凋亡的作用,这可能是其抗肝纤维化的机制之一。
Objective: To observe the effect of losartan on the apoptosis of hepatic stellate cells. Methods: The apoptotic rate of hepatic stellate cells was detected by Annexin V-FITC / PI. The expressions of Bcl-2 and Bax were detected by immunohistochemistry. Results: Double staining assay showed that all concentrations of losartan can promote apoptosis, compared with the control group, the difference was statistically significant (P <0.01). The results of immunohistochemistry showed that losartan decreased the expression of Bcl-2 and the expression of Bax, which was significantly different from the control group (P <0.05). Conclusion: Losartan induces hepatic stellate cells apoptosis, which may be one of the mechanisms of anti-hepatic fibrosis.
