目的研究中药甲宁对自身免疫性甲状腺炎治疗作用。方法用0·05%碘化钠和福氏佐剂抗原乳剂进行造模,2周后加强免疫,以后每周加强免疫1次,至第6周结束[1-3]。正常对照组不做任何处理。各药物组即①~④组免疫后第3周开始灌胃给药,1次/d,均按每100g体质量1ml容积给药,⑤、⑥组大鼠给与等容生理盐水。第10周结束实验。取血(3ml),分离血清冻存(-80℃),测定甲状腺球蛋白抗体(TGAb),甲状腺微粒体抗体(TPOAb)。测量的甲状腺的重量指数。甲状腺组织进行病理检查。结果组织学示雷公藤组及甲宁大、中、小剂量组甲状腺病变程度均较模型组轻,差异有统计学意义(P<0·05)。模型组大鼠重量及重量指数高于正常对照组和各给药组,差异有统计学意义。但各给药组之间差异无统计学意义。模型组大鼠血清TGAb、TPOAb水平明显高于正常组(P<0·001),而各给药组大鼠升高不明显。结论表明甲宁、雷公藤对大鼠自身免疫性甲状腺炎有治疗作用。并且甲宁大剂量组的治疗效果明显优于雷公藤组(P<0·05),中、小剂量组差异无统计学意义。
Objective To study the therapeutic effect of traditional Chinese medicine Jining on autoimmune thyroiditis. Methods The model was prepared with 0.05% sodium iodide and Freund’s adjuvant antigen emulsion, boosted after 2 weeks, then boosted once a week and ended at the end of 6 weeks [1-3]. The normal control group did not do any treatment. Each drug group, that is, 1-4 groups were given intragastric administration 3 weeks after the immunization. Once a day, they were administered at a volume of 1 ml per 100 g of body weight. Rats in groups 5 and 6 were given isotonic saline. The experiment ended in the 10th week. Blood (3 ml) was collected, serum was separated and frozen (-80°C), and thyroglobulin antibody (TGAb) and thyroid microsomal antibody (TPOAb) were measured. Measured thyroid weight index. Thyroid tissue was examined pathologically. Results The histological findings showed that the levels of thyroid lesions in the triptolide group and the large, medium, and small doses of Manning group were lighter than those in the model group (P<0.05). The weight and weight index of rats in the model group were higher than those in the normal control group and each administration group, and the difference was statistically significant. However, there was no statistically significant difference between the groups. The levels of serum TGAb and TPOAb in the model group were significantly higher than those in the normal group (P<0.001), but the rats in each group did not increase significantly. The results showed that Canning and Tripterygium wilfordii can treat autoimmune thyroiditis in rats. And the treatment effect of the high-dose Jining group was significantly better than that of Tripterygium wilfordii group (P<0.05), and there was no significant difference between the medium and low dose groups.
