目的:探讨二烯丙基三硫(DATS)对急性肺损伤小鼠血清IL-18的影响。方法:30只健康昆明小鼠,随机分为5组:(1)ALI组:腹腔注射大肠杆菌脂多糖(LPS);(2)预防组:腹腔注射DATS 7 d后注射LPS;(3)治疗组:腹腔注射LPS后30 min注射DATS;(4)DATS组,腹腔注射DATS 7 d;(5)对照组:腹腔注射等量氯化钠溶液。光镜观察12 h点肺组织形态学改变。测定血清中IL-18在2 h和6 h含量。结果:DATS预防组肺泡间渗出及炎性细胞浸润较ALI组明显减轻。其血清中IL-18含量均明显低于ALI组(P<0.01)。结论:预防性应用DATS可减轻LPS诱导的ALI小鼠肺组织炎症反应,及抑制LPS诱导的小鼠血清中炎性细胞因子IL-18的生成。
Objective: To investigate the effect of diallyl trisulfide (DATS) on serum IL-18 in mice with acute lung injury. Methods: Thirty healthy Kunming mice were randomly divided into 5 groups: (1) ALI group: intraperitoneal injection of E.coli lipopolysaccharide (LPS); (2) prophylaxis group: LPS was injected 7 d after intraperitoneal injection of DATS; (3) Group: DATS was injected 30 min after intraperitoneal injection of LPS; (4) DATS group, intraperitoneal injection of DATS for 7 d; (5) Control group: intraperitoneal injection of equal amount of sodium chloride solution. Light microscopy 12 h point lung morphological changes. Serum IL-18 levels were measured at 2 h and 6 h. RESULTS: Alveolar effusion and inflammatory cell infiltration in the DATS prevention group were significantly reduced compared with those in the ALI group. The serum IL-18 levels were significantly lower than the ALI group (P <0.01). CONCLUSIONS: Prophylactic administration of DATS can attenuate LPS-induced lung inflammation in ALI mice and inhibit the production of IL-18, an inflammatory cytokine induced by LPS in mice.
