[目的]研究人源Her2高表达转基因小鼠的生物学特性,并检测基因KLF4的表达变化。[方法]在以往构建的Her2高表达转基因小鼠稳定遗传体系基础上,传代扩大种群。用PCR方法鉴定子代基因型,绘制体重-周龄曲线。RT-PCR方法检测基因KLF4(FL)及其分型KLF4α的表达。[结果]Her2高表达乳腺癌阳性小鼠体内基因在乳腺癌发生过程中总表达量略微下降;基因的表达量与正常小鼠相比明显增加;然而在乳腺癌阳性小鼠体内KLF4总基因的表达量却呈下降趋势。[结论]该研究能填补KLF4在动物模型水平上研究的空白,使KLF4与乳腺癌发生发展关系的研究更具完整性。
[Objective] To study the biological characteristics of human Her2-overexpression transgenic mice and detect the expression of KLF4 gene. [Method] On the basis of the stable genetic system of Her2 overexpressing transgenic mice constructed in the past, the expanded population was subcultured. The genotypes of offspring were identified by PCR and the body weight - week old curve was drawn. RT-PCR was used to detect the expression of KLF4 (FL) and KLF4α. [Results] The expression of gene in Her2 overexpressing breast cancer positive mice decreased slightly during the development of breast cancer. The gene expression of Her2 overexpression increased significantly compared with that of normal mice. However, in the positive breast cancer mice, the total KLF4 gene The amount of expression showed a downward trend. [Conclusion] The study can fill the blank of KLF4 research in animal models and make the study on the relationship between KLF4 and breast cancer more complete.
