目的:研究X线对Tca8113细胞端粒重复序列结合因子2蛋白的影响。方法:采用Western Blot检测X线照射前后端粒重复序列结合因子2蛋白的含量变化,采用AnnexinⅤ-FITC/PI联合染色检测细胞早期凋亡。结果:与照射前相比,细胞早期凋亡率显著升高(P<0.05);经不同剂量X线照射后0.5 h,各组端粒重复序列结合因子2蛋白含量均上升(P<0.05),1 h后,2 Gy组的TRF2蛋白含量降至与未照射时水平相当,3 h和5 h时降至比未照射时更低(P<0.05),其它照射剂量组的TRF2蛋白在照射后1 h时均已降至不可测得的水平。结论:端粒重复序列结合因子2被激活,端粒重复序列结合因子2蛋白含量应激性地短暂升高是经X线照射后的早期事件。
Objective: To study the effect of X-ray on telomeric repeat binding protein 2 in Tca8113 cells. Methods: The changes of telomeric repeat binding protein 2 protein content before and after X-ray irradiation were detected by Western Blot. The cell apoptosis was detected by AnnexinⅤ-FITC / PI staining. Results: Compared with the pre-irradiation, the early apoptotic rate of cells was significantly increased (P <0.05). After 0.5 h of irradiation, the telomeric repeat binding protein 2 content increased (P <0.05) After 1 h, TRF2 protein content in 2 Gy group was reduced to the same level as that of non-irradiated group, but decreased to that of non-irradiated group at 3 h and 5 h (P <0.05) After 1 h have dropped to an unmeasurable level. CONCLUSIONS: The telomeric repeat binding factor 2 is activated and the transient, transiently elevated telomeric repeat binding factor 2 protein content is an early event after X-ray irradiation.
