目的:在蛋白水平和mRNA水平定量研究肽基精氨酸脱亚氨酶(PADI4)在各种不同类型肿瘤及对照组织中的表达,并分析食管鳞癌组织中表达量与临床相关因素的关系。方法:免疫组化和免疫荧光的方法定性、定量研究PADI4在多种肿瘤中的表达。蛋白质印迹法和Real-ti mePCR定量分析PADI4蛋白水平和mRNA水平上在各种肿瘤中表达水平。结果:PADI4在食管癌(14例,阳性率100%)、胃癌(5例,80%)、结肠癌(5例,60%)、直肠癌(5例,80%)、肾癌(5例,100%)、肺癌(5例,100%)组织中的表达量明显高于癌旁组织,差异有统计学意义,P<0.05。子宫癌(5例,阳性率80%)、乳腺癌(5例,80%)和卵巢癌(5例,100%)中的表达量分别高于子宫肌瘤(5例,20%)、乳腺增生(5例,20%)和卵巢囊腺瘤(5例,0),差异有统计学意义,P<0.05。蛋白质印迹法和Real-ti me PCR分别在蛋白水平和mRNA水平上证明以上结果。在食管鳞癌(139例,阳性率98.56%)、食管腺癌(16例,87.50%)和食管小细胞未分化癌(12例,91.67%)中PADI4高表达,正常食管组织(16例,0)中不表达。在食管鳞癌组织中,PADI4的表达与病理分级呈正相关,P<0.05。结论:PADI4在恶性肿瘤中大量表达,且在食管鳞癌组织中表达量与肿瘤的病变程度正相关,该酶可能在恶性肿瘤的发生中起重要作用。
OBJECTIVE: To quantitatively study the expression of peptidyl arginine deiminase (PADI4) in various types of tumors and control tissues at the protein and mRNA levels, and to analyze the relationship between the expression of PADI4 and clinical related factors in esophageal squamous cell carcinoma . Methods: Immunohistochemistry and immunofluorescence were used to qualitatively and quantitatively study the expression of PADI4 in various tumors. Western blot and Real-ti mePCR were used to quantitatively analyze the levels of PADI4 protein and mRNA expression in various tumors. Results: The positive rate of PADI4 in esophageal cancer (14 cases, 100%), gastric cancer (5 cases, 80%), colon cancer (5 cases, 60%), rectal cancer (5 cases, 80% , 100%) and lung cancer (5 cases, 100%), the difference was statistically significant (P <0.05). The expression of uterine cancer (5 cases, 80%), breast cancer (5 cases, 80%) and ovarian cancer (5 cases, 100% Hyperplasia (5 cases, 20%) and ovarian cystadenoma (5 cases, 0), the difference was statistically significant, P <0.05. Western blotting and Real-ti me PCR demonstrated the above results at the protein and mRNA levels, respectively. PADI4 was highly expressed in esophageal squamous cell carcinoma (139 cases, positive rate 98.56%), esophageal adenocarcinoma (16 cases, 87.50%) and esophageal small cell undifferentiated carcinoma (12 cases, 91.67% 0) is not expressed. In esophageal squamous cell carcinoma, PADI4 expression was positively correlated with pathological grade (P <0.05). CONCLUSION: PADI4 is highly expressed in malignant tumors, and its expression in esophageal squamous cell carcinoma is positively correlated with the degree of tumorigenesis. This enzyme may play an important role in the carcinogenesis of malignant tumors.
