目的分析动脉粥样硬化斑块与龈下菌斑中牙周致病菌的关系。方法选取2014年8月—2015年10月于新疆医科大学第一附属医院心脏外科行冠状动脉旁路移植术的冠心病并牙周炎患者86例,收集其动脉粥样硬化斑块及龈下菌斑。采用Chelex-100法及聚合酶链反应(PCR)提取动脉粥样硬化斑块及龈下菌斑中福赛斯坦纳菌(Tf)DNA,计算动脉粥样硬化斑块和龈下菌斑中Tf DNA拷贝数,动脉粥样硬化斑块与龈下菌斑中Tf占比的相关性分析采用Pearson相关性分析。结果动脉粥样硬化斑块和龈下菌斑中Tf阳性检出率分别为60.71%、80.21%。动脉粥样硬化斑块和龈下菌斑中Tf DNA测序结果符合Genbank数据库相应细菌标准序列,同源性达99%。动脉粥样硬化斑块中Tf DNA拷贝数为(2.24×10~2±1.52×10~2)拷贝/μl,Tf占比为(0.28±0.13)%;龈下菌斑中Tf DNA拷贝数为(2.06×10~3±1.18×10~4)拷贝/μl,Tf占比为(0.35±0.22)%;动脉粥样硬化斑块和龈下菌斑中Tf占比比较,差异有统计学意义(P<0.05)。Pearson相关性分析结果显示,动脉粥样硬化斑块与龈下菌斑中Tf占比呈正相关(r=0.498,P<0.05)。结论动脉粥样硬化斑块和龈下菌斑中Tf阳性检出率均较高,动脉粥样硬化斑块和龈下菌斑中Tf具有同源性,且动脉粥样硬化斑块与龈下菌斑中Tf相关。
Objective To analyze the relationship between atherosclerotic plaque and periodontal pathogens in subgingival plaque. Methods From August 2014 to October 2015, 86 patients with coronary artery disease and periodontitis underwent coronary artery bypass grafting in the First Affiliated Hospital of Xinjiang Medical University were enrolled. Their atherosclerotic plaques and subgingival Plaque. The Tf DNA of atherosclerotic plaque and subgingival plaque was extracted by Chelex-100 method and polymerase chain reaction (PCR) to calculate the Tf of atherosclerotic plaque and subgingival plaque DNA copy number, atherosclerotic plaque and subgingival plaque in the proportion of Tf correlation analysis using Pearson correlation analysis. Results The positive rates of Tf in atherosclerotic plaque and subgingival plaque were 60.71% and 80.21%, respectively. The results of Tf DNA sequencing in atherosclerotic plaques and subgingival plaques were in accordance with the corresponding bacterial sequences in the Genbank database, with a homology of 99%. The Tf DNA copy number was (2.24 × 10 ~ 2 ± 1.52 × 10 ~ 2) copies / μl in atherosclerotic plaque, and the Tf proportion was (0.28 ± 0.13)%. The copy number of Tf DNA in subgingival plaque was (2.06 × 10 ~ 3 ± 1.18 × 10 ~ 4) copies / μl, Tf accounted for (0.35 ± 0.22)%; The difference of Tf in atherosclerotic plaque and subgingival plaque was statistically significant (P <0.05). Pearson correlation analysis showed that atherosclerotic plaque was positively correlated with the proportion of Tf in subgingival plaque (r = 0.498, P <0.05). Conclusions The positive detection rate of Tf in atherosclerotic plaque and subgingival plaque is high, Tf in atherosclerotic plaque and subgingival plaque are homologous, and atherosclerotic plaque and subgingival plaque Tf related to plaque.
