目的 研究 3种穿心草 口山酮 (xanthone ,Xan)对心肌线粒体氧化损伤的保护作用。方法 采用抗坏血酸和硫酸亚铁为线粒体损伤诱导剂 ,探讨Xan对线粒体的膨胀、脂质过氧化及膜流动性的影响。结果 3种Xan不同程度地减少受伤线粒体的膨胀作用及阻止脂质过氧化的形成 ,增加膜脂质流动性 ,维护线粒体结构和功能的完整 ,作用强度依次为Xan Ⅰ >Xan Ⅲ >Xan Ⅱ。结论 3种Xan对心肌线粒体氧化损伤均有不同程度的保护作用 ,作用机制可能与其清除自由基、抗脂质过氧化作用有关 ,而 口山酮活性强弱可能与其苯环上酚羟基和甲氧基的数量及位置相关
Objective To study the protective effect of xanthone (Xan) against oxidative damage of myocardial mitochondria. Methods Ascorbic acid and ferrous sulfate were used as inducers of mitochondrial damage to investigate the effects of Xan on mitochondria swelling, lipid peroxidation and membrane fluidity. Results Xan reduced the mitochondria swelling and the formation of lipid peroxidation, increased the membrane lipid fluidity and maintained the integrity of mitochondrial structure and function in order of Xan Ⅰ> Xan Ⅲ> Xan Ⅱ. Conclusions The three kinds of Xan have protective effects on myocardial mitochondrial oxidative damage in varying degrees, and the mechanism may be related to the scavenging of free radicals and anti-lipid peroxidation.
