目的 研究人垂体腺瘤中的细胞凋亡和凋亡相关基因 (P53、Bcl 2、Bax)表达蛋白的情况,分析其和临床特征的相关性。方法 采用流式细胞仪检测 21例人垂体腺瘤中的凋亡和增殖细胞比例,采用TUNEL法检测 14例细胞凋亡,同时应用免疫组化技术检测 17例细胞增殖相关抗原(PCNA),凋亡相关基因P53、Bcl 2和 13例Bax表达的蛋白产物。结果 TUNEL法在 14例垂体腺瘤中 12例检测到细胞凋亡 (占 85 71% ), 其凋亡细胞比例 0 1% ~8 0%,流式细胞仪检测 21例中凋亡细胞比例为 1 77%±2 12%,两者和垂体腺瘤临床特征无明显相关性。凋亡相关基因中Bax阳性率高(16 /17, 92 31% ),表达强度高,P53有 2例阳性, Bcl 2仅 1例阳性。Bax表达和细胞增殖活性相关显著。结论 人垂体腺瘤中存在细胞凋亡,垂体腺瘤中Bax高表达,Bcl 2 /Bax比值 <1 0,这一细胞内微环境有利于细胞凋亡的存在。垂体腺瘤的凋亡活性和其临床特征 (如增殖活性,侵袭性等 )无明显相关性。
Objective To study the expression of apoptosis-related genes (P53, Bcl 2, Bax) in pituitary adenomas and to analyze their correlation with clinical features. Methods Flow cytometry was used to detect the percentage of apoptotic cells and proliferating cells in 21 cases of human pituitary adenoma. TUNEL assay was used to detect the apoptosis of 14 cases. Meanwhile, immunohistochemistry was used to detect the expression of PCNA, Related gene P53, Bcl 2 and 13 cases of Bax protein product. Results TUNEL assay showed that apoptotic cells were detected in 12 of 14 pituitary adenomas (accounting for 85 71%), the percentage of apoptotic cells was 0 1% ~ 80%, and the percentage of apoptotic cells in flow cytometry was 21 1 77% ± 2 12%. There was no significant correlation between them and the clinical features of pituitary adenomas. The positive rate of Bax in apoptosis-related genes (16/17, 92 31%) was high, the expression of Bax was high, 2 cases were positive for P53, and 1 case was positive for Bcl 2. Bax expression and cell proliferation were significantly correlated. Conclusions There is apoptosis in human pituitary adenoma. The expression of Bax in pituitary adenoma is high, and the ratio of Bcl 2 / Bax is less than 10. This intracellular microenvironment favors the existence of apoptosis. Pituitary adenoma apoptosis and its clinical features (such as proliferation activity, invasiveness, etc.) no significant correlation.
