目的建立高效液相色谱法测定新疆鼠尾草抗糖尿病并发症有效部位中丹酚酸K含量的方法。方法色谱柱为SHIMADZU SHIM-PACK VP-ODS C18(250 mm×4.6 mm,5μm);流动相为0.2%磷酸-甲醇(65∶35,V/V,调pH值为3.0);流速为0.8 ml.min-1;检测波长为320 nm;柱温为30℃。结果丹酚酸K与其它组分的分离效果良好,且在0.1~0.35μg范围内呈良好的线性关系(r≥0.999 9),平均加样回收率为98.11%(RSD=1.87%,n=9)。结论该测定方法简便、准确、可行,为新疆鼠尾草有效部位质量控制和新药开发提供科学依据。
Objective To establish a method for the determination of the content of salvianolic acid K in the effective part of anti-diabetic complications of sage by high performance liquid chromatography. METHODS The chromatographic column was SHIMADZU SHIM-PACK VP-ODS C18 (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of 0.2% phosphoric acid-methanol (65:35 V / V, adjusted to pH 3.0) .min-1; detection wavelength was 320 nm; column temperature was 30 ℃. Results The separation of salvianolic acid K from other components was good and the linearity was within the range of 0.1 ~ 0.35 μg (r ≥ 0.99 9). The average recovery was 98.11% (RSD = 1.87%, n = 9). Conclusion The method is simple, accurate and feasible, providing a scientific basis for the quality control and new drug development of the effective part of Sage.
