基质细胞衍生因子1对内源性神经干细胞的趋化迁移作用

背景:目前研究表明,基质细胞衍生因子1在参与趋化迁移内源性神经干细胞中起着非常重要的作用,但其具体迁移机制尚不明确。目的:观察外源性基质细胞衍生因子1对大鼠内源性神经干细胞的趋化迁移作用及海马区神经干细胞的激活增殖情况。方法:通过向SD大鼠海马区上大脑皮质内注射外源性基质细胞衍生因子1(注射量为5μL,质量浓度为500μg/L)建立动物模型,于3,7,14,21 d后灌注取脑,通过石蜡切片免疫组织化学检测大鼠皮质内注射区及海马区nestin阳性细胞表达情况,并设实验对照组与空白对照组。结果与结论:石蜡切片免疫组织化学显示:实验组注射区周围及海马区nestin表达阳性细胞的数量随时间推移逐渐增多,3,7 d时注射区及海马区nestin表达阳性细胞少量表达,14 d时注射区及海马区nestin表达阳性细胞进一步增多,并向注射区形成明显的迁移趋势,21 d时注射区及海马区nestin表达阳性细胞更多。实验对照组及空白实验组未见上述表现。结果表明外源性基质细胞衍生因子1可能诱导海马区神经干细胞的增殖分化,参与内源性神经干细胞的趋化迁移过程。 BACKGROUND: Current research shows that stromal cell-derived factor 1 plays a very important role in the chemotaxis of endogenous neural stem cells. However, its specific migration mechanism is not yet clear. OBJECTIVE: To observe the chemotaxis of exogenous stromal cell-derived factor 1 to rat endogenous neural stem cells and the activation and proliferation of neural stem cells in hippocampus. Methods: Animal models were established by injection of exogenous stromal cell-derived factor 1 (5 μL, mass concentration 500 μg / L) into the cerebral cortex of hippocampus of SD rats. After 3, 7, 14 and 21 days, The brain tissues were collected and the expression of nestin positive cells in the cortex injection area and the hippocampus area were detected by paraffin section immunohistochemistry. The experimental control group and the blank control group were also established. RESULTS AND CONCLUSION: Immunohistochemistry of paraffin sections showed that the number of nestin positive cells around the injection area and hippocampus in experimental group gradually increased over time. At 3 and 7 days, the expression of nestin positive cells in injection area and hippocampus area was small, The number of nestin positive cells in the injection and hippocampus increased further, and a significant migration tendency was found in the injection zone. At 21 d, the number of nestin positive cells in the injection zone and the hippocampus area was more. Experimental control group and blank experimental group did not show the above performance. The results show that exogenous stromal cell-derived factor 1 may induce the proliferation and differentiation of neural stem cells in the hippocampus and participate in the chemotactic migration of endogenous neural stem cells.
