我们在做免疫组化SABC法过程中,采用一般家用微波炉抗原修复,整个实验过程如下:(1)石蜡切片经二甲苯脱蜡至蒸馏水。(2)抗原修复:中等火力,5min×2,在微波炉中静置15min,然后置于室温,冷至50℃以下。(3)以下步骤参照试剂说明书。 在整个实验过程中,关键是第2步抗原修复,我们采用2个5min,中等火力,切片在枸椽酸中沸腾,结果良好;如果在第2个5min抗原修复时,液体不沸腾,抗原得不到充分暴露,那么,在显色反应中标本根本不显色,此时,可再经微波处理5min至沸腾。仍可取得良好效果。
