目的:采用体外炎症模型研究瓜子金有效部位群抗炎作用机制。方法:建立脂多糖(LPS)诱导巨噬细胞系RAW264.7细胞体外炎症模型,研究不同浓度瓜子金有效部位群对一氧化氮(NO)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的释放及对TNF-α和IL-6 mRNA表达的影响。结果:瓜子金有效部位群能显著降低LPS诱导的RAW264.7细胞NO,IL-6及TNF-α的释放,显著降低TNF-α及IL-6 mRNA的表达。结论:瓜子金有效部位群抗炎作用与其减少巨噬细胞炎症介质的生成与释放及降低TNF-α和IL-6 mRNA的表达密切相关。
OBJECTIVE: To study the anti-inflammatory mechanism of melon seeds gold effective part in vitro using in vitro inflammation model. Methods: Lipopolysaccharide (LPS) -induced macrophage cell line RAW264.7 cells in vitro inflammatory model to study the effect of different concentrations of melon gold effective sites on nitric oxide (NO), tumor necrosis factor-α (TNF-α), interleukin 6 (IL-6) release and its effect on the expression of TNF-α and IL-6 mRNA. Results: The effective part of melon seeds could significantly reduce the release of NO, IL-6 and TNF-α in LPS-induced RAW264.7 cells and significantly decrease the expression of TNF-α and IL-6 mRNA. Conclusion: The anti-inflammatory effect of the effective parts of melon seeds gold is closely related to the formation and release of inflammatory mediators and the decrease of the expression of TNF-α and IL-6 mRNA in macrophages.
