抗日本血吸虫病双价疫苗pVAX1/SjCB?hIL-18的构建及其动物保护性研究

目的探讨抗日本血吸虫病双价疫苗pVAX1/SjCB·hIL-18的构建以及对小鼠的保护作用。方法构建pVAX1/SjCB、pVAX1/hIL-18单价疫苗以及pVAX1/SjCB·hIL-18双价疫苗,选择100只无特定病原体的雌性小鼠,随机分为pVAX1/SjCB·hIL-18组、pVAX1/SjCB组、pVAX1/hIL-18组、pVAX1组和对照组各20只。分别于第0、2、4周在小鼠的背部皮下注射疫苗100μg,pVAX1组和对照组分别注射pVAX1和生理盐水。比较各种小鼠间的减虫率和减卵率。结果 pVAX1/SjCB·hIL-18组、pVAX1/SjCB组、pVAX1/hIL-18组小鼠的减虫率以及减卵率均明显高于pVAX1组和对照组小鼠(P<0.05)。pVAX1/SjCB·hIL-18组小鼠的减虫率以及减卵率明显高于pVAX1/SjCB组和pVAX1/hIL-18组小鼠(P<0.05)。结论 pVAX1/SjCB、pVAX1/hIL-18单价疫苗以及pVAX1/SjCB·hIL-18双价疫苗均对日本血吸血虫病具有良好的保护效果,其中pVAX1/SjCB·hIL-18双价疫苗的保护作用最佳,值得临床推广。 Objective To investigate the construction of bivalent vaccine against Japanese schistosomiasis pVAX1 / SjCB · hIL-18 and its protective effect on mice. Methods The pVAX1 / SjCB, pVAX1 / hIL-18 monovalent vaccine and pVAX1 / SjCB · hIL-18 bivalent vaccine were constructed and 100 female mice without specific pathogen were selected and randomly divided into pVAX1 / SjCB · hIL- SjCB group, pVAX1 / hIL-18 group, pVAX1 group and control group. At the 0th, 2nd, 4th week respectively, the mice were injected subcutaneously with 100μg vaccine. The pVAX1 group and the control group were injected with pVAX1 and saline respectively. Compare the worm reduction and egg reduction rates among various mice. Results The worm reduction rate and the rate of egg reduction in pVAX1 / SjCB · hIL-18, pVAX1 / SjCB and pVAX1 / hIL-18 groups were significantly higher than those in pVAX1 and control groups (P <0.05). The worm reduction rate and the rate of egg reduction in pVAX1 / SjCB · hIL-18 group were significantly higher than those in pVAX1 / SjCB group and pVAX1 / hIL-18 group (P <0.05). Conclusion Both pVAX1 / SjCB, pVAX1 / hIL-18 monovalent vaccine and pVAX1 / SjCB · hIL-18 bivalent vaccine have a good protective effect against Japanese bloodsucker disease.
