目的:利用去卵巢大鼠骨质疏松模型,观察骨碎补总黄酮对血清中肿瘤坏死因子α(TNF-α)、白介素1(IL-1)、前列腺素E2(PE2)和5羟色胺(5-HT)的表达和对大鼠痛阈的影响。方法:36只雌性大鼠随机分为3组(假手术组、去卵巢组和药物处理组),造模成功后,药物处理组持续给予骨碎补总黄酮,采用ELISA检测大鼠血清中TNF-α、IL-1、PE2和5-HT的表达变化,采用X线骨密度检测仪测定大鼠骨密度;采用von-Frey纤毛检测法观察其不同时间点的镇痛作用。结果:骨碎补总黄酮对去卵巢骨质疏松症导致的骨密度下降有明显抑制作用,且明显抑制大鼠血清中TNF-α、IL-1、PE2和5-HT的表达,显著提高去卵巢骨质疏松大鼠的痛阈。结论:骨碎补总黄酮能明显提高去卵巢大鼠的骨密度,并可抑制血清中疼痛介质的表达,具有镇痛作用。
OBJECTIVE: To observe the effects of total flavonoids of Rhizoma Drynariae on serum levels of tumor necrosis factor-α (TNF-α), interleukin-1 (IL-1), prostaglandin E2 (PE2) and serotonin 5 -HT) and its effect on pain threshold in rats. Methods: Thirty-six female rats were randomly divided into three groups (sham-operated group, ovariectomized group and drug-treated group). After successful modeling, the total flavonoids of Rhizoma Drynariae were given to the drug- The expressions of IL-1, IL-1, PE2 and 5-HT were measured by radioimmunoassay. The bone mineral density (BMD) was measured by X-ray absorptiometry. The von-Frey cilia were used to observe the analgesic effect at different time points. Results: Total Flavonoids of Rhizoma Drynariae significantly inhibited the decrease of bone mineral density induced by osteoporosis in ovariectomized rats and significantly inhibited the expression of TNF-α, IL-1, PE2 and 5-HT in serum Pain threshold in ovarian osteoporosis rats. Conclusion: Total Flavonoids of Rhizoma Drynariae can significantly increase bone mineral density in ovariectomized rats and inhibit the expression of pain mediators in serum, which has the analgesic effect.
