目的:建立反相高效液相色谱法测定比格犬血浆中格列美脲(GM)的浓度。方法:色谱柱为Agilent Eclipse XDB-C8,柱温为30℃,流动相为乙腈-甲醇-10mmol·L-1磷酸二氢钾(65∶40∶95,pH7.2),流速为1.0mL·min-1,检测波长为227nm。以吉非罗齐为内标,采用液-液萃取法预处理血浆样品,测定6只比格犬单剂量给予GM舌下喷雾剂(2mg)后不同时间点的血药浓度。结果:GM检测浓度线性范围为50~2500ng·mL-1,最低检出限为20ng·mL-1,日内和日间RSD均小于15%,萃取回收率为71.27%~103.5%,方法回收率为87.97%~98.95%。结论:本方法样品处理简单,灵敏度高,适合分析生物样品中的GM。
Objective: To establish a method for the determination of glimepiride (GM) in Beagle dog plasma by reversed-phase high performance liquid chromatography. Methods: The column was Agilent Eclipse XDB-C8. The mobile phase consisted of acetonitrile-methanol-10mmol·L-1 potassium phosphate monobasic (65:40:95, pH7.2) at a flow rate of 1.0mL · min-1, detection wavelength of 227nm. Taking gemfibrozil as internal standard, the plasma samples were pretreated by liquid-liquid extraction and the plasma concentrations of 6 beagle dogs at different time points after single-dose administration of GM sublingual spray (2 mg) were determined. Results: The linear range of GM was 50-2500 ng · mL-1, the lowest detection limit was 20 ng · mL-1. The RSD of intra-day and inter-day was less than 15% and the recovery of extraction was 71.27% -103.5%. The method recovery For the 87.97% ~ 98.95%. Conclusion: The method is simple, sensitive and suitable for the analysis of GM in biological samples.
