目的:研究槲皮素对人肺腺癌细胞株A549和小鼠Lewis肺癌移植瘤细胞增殖的抑制作用,并探讨其可能的 机制。 方法:①以不同剂量的槲皮素作用于人肺腺癌细胞株A549,用噻唑蓝(MTT)试验观察槲皮素对人肺癌细 胞株增殖的影响。②将移植有Lewis肺癌的ICR小鼠随机分为模型对照组、槲皮素组及顺铂组,均采取腹腔内注射 方式给药。给药结束后,测定各组小鼠的瘤重,计算抑瘤率;用免疫组化方法测定肿瘤的增殖细胞核抗原(PCNA) 表达水平。 结果:①槲皮素对A549细胞增殖有明显的抑制作用,对细胞增殖的抑制率呈剂量效应关系。②槲皮 素对小鼠Lewis肺癌移植瘤生长有明显的抑制作用,并显著下调小鼠Lewis肺癌移植瘤的PCNA表达水平。 结 论:槲皮素对人肺腺癌细胞A549和小鼠Lewis肺癌移植瘤细胞增殖有明显的抑制作用,其机制可能与抑制PCNA 蛋白的表达有关。
Objective: To study the inhibitory effect of quercetin on the proliferation of human lung adenocarcinoma cell line A549 and Lewis lung carcinoma xenograft in mice and to explore its possible mechanism. Methods: ① The human lung adenocarcinoma cell line A549 was treated with different doses of quercetin, and the effect of quercetin on the proliferation of human lung cancer cell lines was observed by MTT assay. The ICR mice transplanted with Lewis lung cancer were randomly divided into model control group, quercetin group and cisplatin group, and were given intraperitoneal injection. After administration, the tumor weight of the mice in each group was measured to calculate the tumor inhibition rate; and the expression level of proliferating cell nuclear antigen (PCNA) in the tumor was determined by immunohistochemistry. Results: ① Quercetin significantly inhibited the proliferation of A549 cells and inhibited the proliferation of cells in a dose-dependent manner. Quercetin significantly inhibited the growth of transplanted Lewis lung carcinoma in mice and down-regulated the expression of PCNA in Lewis lung carcinoma xenografts in mice. Conclusion: Quercetin can significantly inhibit the proliferation of human lung adenocarcinoma A549 cells and Lewis lung carcinoma xenografts in mice, and its mechanism may be related to the inhibition of the expression of PCNA protein.
