以邻苯二醛(OPA)作柱后衍生化试剂,可快速且有效地用HPLC 法分离新霉素A、B 及C(下简称A,B 及C)和测定。柱后反应系统由T 形结构混合器及螺旋状不锈钢毛细管反应管组成;OPA 试液新鲜配制,含60mg/lOPA 和0.2%(V/V)2-巯基乙醇的0.2MpH 9.3四硼酸钠缓冲液。本法以滤光电荧光计检测,在流动相泵与进样阀间装置一卫柱(20×4.6 mm,id)以10 μm Lichrosrb RP-18填充。
With phthalaldehyde (OPA) as a post-column derivatization reagent, neomycin A, B and C (hereinafter referred to as A, B and C) can be rapidly and efficiently separated by HPLC and determined. The post-column reaction system consists of a T-shaped structure mixer and a helical stainless steel capillary reaction tube. The OPA test solution was freshly prepared with 0.2 M pH 9.3 sodium tetraborate buffer solution containing 60 mg / l OPA and 0.2% (v / v) 2- mercaptoethanol .
