本文介绍实验性矽肺研究的透射电镜制样方法。标本采用戊二醛-多聚甲醛混合固定液固定,在负压中进一步固定;延长脱水与浸透时间,并辅以机械振荡;选用大角度玻璃刀及较慢速度切片等措施,获得较为满意的结果。 This article describes the experimental method of transmission electron microscopy for silicosis research. The specimens were fixed with glutaraldehyde-paraformaldehyde mixed fixative and further fixed in negative pressure. The dehydration and soaking time were prolonged, supplemented by mechanical oscillations. Large-angle glass knife and slow speed slicing were selected to obtain satisfactory results result.
