目的:研究祛斑汤对Ha Cat细胞、黑素细胞的抑制作用及对SCF/C-kit蛋白表达水平的影响,探讨SCF/C-kit信号通路与黄褐斑发病机制的相关性。方法:MTT法观察0、5、15、25、50、100 mg/m L浓度祛斑汤对Ha Cat细胞、黑素细胞的抑制情况,选取最佳抑制浓度;根据最佳抑制浓度,将Hacat细胞分为1640培养基组、0.03 J/cm2UVB照射组、15 mg/m L祛斑汤组、0.03 J/cm2UVB照射+15 mg/m L祛斑汤组,黑素细胞分为Mel M-2培养基组、0.03 J/cm2UVB照射组、25 mg/m L祛斑汤组、0.03 J/cm2UVB照射+25 mg/m L祛斑汤组;Western blot法检测各组Hacat细胞SCF蛋白表达水平,各组黑素细胞的C-kit受体蛋白表达水平。结果:祛斑汤呈浓度依赖性抑制Hacat细胞/黑素细胞增殖,15 mg/m L时对Hacat细胞的抑制效应达到峰值(P<0.01),25 mg/m L时对黑素细胞的抑制效应达到峰值(P<0.01);祛斑汤抑制Hacat细胞SCF蛋白的表达和黑素细胞C-kit受体蛋白的表达,并下调UVB对Ha Cat细胞、黑素细胞SCF、C-kit蛋白表达的促进作用。结论:祛斑汤能有效抑制Hacat细胞、黑素细胞的增殖,可能与抑制SCF/C-kit信号途径的SCF、C-kit蛋白表达有关。
Objective: To study the inhibitory effect of Quban Decoction on Ha Cat cells and melanocytes and its effect on the expression of SCF / C-kit protein and to explore the correlation between SCF / C-kit signaling pathway and the pathogenesis of melasma. The inhibitory effect of Quban decoction on Hacat cells reached a peak at 15 mg / m L (P <0.01), and the inhibitory effect on melanocytes at 25 mg / m L (P <0.01). Freckle decoction could inhibit the expression of SCF protein and melanocyte C-kit receptor protein in Hacat cells and decrease the expression of SCF and C-kit protein in Ha Cat cells and melanocytes effect. Conclusion: Quban Decoction can effectively inhibit the proliferation of Hacat cells and melanocytes, which may be related to the inhibition of SCF and C-kit protein expression in SCF / C-kit signaling pathway.
