目的以鼠麴草的叶为材料,研究鼠麴草DNA的提取方法及对其进行随机扩增多态性DNA(RAPD)的分析。方法采用略改进的十六烷基三甲基溴化铵(CTAB)法、高盐低pH法、木本法和十二烷基硫酸钠(SDS)法分别提取鼠麴草叶的基因组DNA。通过RAPD分析所提取的DNA,比较所用的提取方法。结果CTAB法得到DNA的A260/A280为1.937,浓度为992.25μg/ml,优于其他3种方法。结论CTAB法是4种方法中最佳的方法。
Objective To study the method of extracting DNA from Morinda officinalis and its analysis of random amplified polymorphic DNA (RAPD) using leaves of Morinda citrifolia as material. Methods The genomic DNA of leaves of Morinda citrifolia L. was extracted by slightly modified cetyltrimethylammonium bromide (CTAB) method, high salt low pH method, woody method and sodium dodecyl sulfate (SDS) method. The extracted DNA was compared by RAPD analysis and the extraction method used was compared. Results The DNA A260/A280 was 1.937 and the concentration was 992.25 μg/ml by CTAB method, which was superior to the other three methods. Conclusion The CTAB method is the best one of the four methods.
