目的观察房间隔缺损(ASD)、室间隔缺损(VSD)介入封堵术前后血小板活化状态的变化。方法选择成功进行ASD、VSD介入封堵术的患者32例(其中ASD16例、VSD16例),分别于术前、术后即刻、术后第4天抽静脉血并ELISA法测定血小板α颗粒膜蛋白(GMP-140)浓度。结果(1)ASD、VSD患者介入封堵术后血浆GMP-140浓度明显升高(P<0.01),术后第4天血浆GMP-140浓度亦有所下降(P<0.05),VSD患者介入封堵术后GMP-140浓度升高较ASD患者更明显(P<0.01);(2)ASD和VSD患者介入封堵术后即刻GMP-140浓度均与手术时间成正相关。结论(1)ASD、VSD封堵术后均存在血小板活化状态,二者相比VSD封堵术后血小板活化更明显;(2)ASD和VSD患者介入封堵术后即刻GMP-140浓度均与手术时间成正相关。
Objective To observe the changes of platelet activation before and after atrial septal defect (VSD) and atrial septal defect (ASD). Methods Thirty-two patients (16 ASDs and 16 VSDs) were successfully treated with ASD and VSD. Venous blood was collected preoperatively and immediately after operation and venous blood was collected on day 4 postoperatively. ELISA was used to determine the expression of platelet α-granule membrane protein (GMP-140) concentration. Results (1) Plasma GMP-140 concentration in patients with ASD and VSD was significantly higher after interventional closure (P <0.01), and plasma GMP-140 concentration also decreased on the 4th day after operation (P <0.05) The concentration of GMP-140 in patients with ASD was significantly higher than that in patients with ASD (P <0.01). (2) The concentration of GMP-140 in patients with ASD and VSD was positively correlated with the time of operation. CONCLUSIONS: (1) Platelet activation exists after ASD and VSD occlusion, both of which are more obvious than VSD occlusion. (2) GMP-140 concentration in patients with ASD and VSD is Surgery time is positively correlated.
