目的从能量代谢角度探讨桂枝甘草汤“辛甘化阳”配伍原理的内涵。方法 SPF级SD大鼠饲养于低温(4~6℃)环境,并分别于给药第15天、30天,每组各处理10只动物,分别采集相同部位心肌组织0.1 g左右,精密称重,加9倍体积的PBS缓冲液制成浓度为10%的组织匀浆液,离心,分离上清液,采用Elisa法检测PGC1α、CS、α-KGDHC含量。结果随给药时间的延长,桂枝甘草汤能持续增高大鼠心肌PGC1α、CS、α-KGDHC含量(P<0.01,P<0.05),并且其作用是通过桂枝、甘草配伍共同实现的。结论桂枝甘草汤“辛甘化阳”配伍原理的内涵与调节心肌能量代谢,增高低温环境大鼠心肌PGC1α、CS、α-KGDHC含量有关。
Objective To explore the connotation of compatibility principle of Guizhi Gancao Decoction “Xin Gan Yang Yang” from the perspective of energy metabolism. Methods SPF SD rats were housed in low temperature (4 ~ 6 ℃) environment and were treated on the 15th day and the 30th day respectively. Ten animals in each group were treated with about 0.1 g of myocardial tissue collected at the same site. Precision weighing , Add 9 times the volume of PBS buffer to make the tissue homogenate with the concentration of 10%, centrifuge, separate the supernatant, measure the content of PGC1α, CS, α-KGDHC by Elisa method. Results With the prolonging of administration time, Guizhi Gancao Decoction could continuously increase the contents of PGC1α, CS and α-KGDHC in myocardium of rats (P <0.01, P <0.05), and its effect was achieved through the combination of Guizhi and Licorice. Conclusions The connotation of compatibility principle of Guizhi Gancao Decoction is related to regulating myocardial energy metabolism and increasing PGC1α, CS and α-KGDHC contents in myocardium of hypothermic rats.
