目的观察亚硒酸钠对肺癌细胞株A549细胞株自噬相关蛋白Beclin1、LC3的作用。方法将人肺癌A549细胞株分为0、0.5、2.5、5.0、8.0μmol/L亚硒酸钠共5组,用不同浓度的亚硒酸钠培养液培养24h、48h后,用免疫细胞化学染色和Western blot印迹法检测亚硒酸钠对自噬相关蛋白Beclin1、LC3表达的影响。结果人肺癌A549细胞株用不同浓度的亚硒酸钠培养24h,48h后,细胞数量逐渐减少,细胞肿胀,变大变圆,染色质凝集、核固缩并断裂。免疫细胞化学和Western blot印迹法结果均显示:随着亚硒酸钠浓度增加,肺癌A549细胞中Beclin1和LC3蛋白免疫细胞化学染色的平均光密度(MOD)有明显提高(P<0.05)。结论亚硒酸钠可以抑制肺癌A549细胞生长,诱导肺癌A549细胞自噬。
Objective To observe the effect of sodium selenite on autophagy-related proteins Beclin1 and LC3 in lung cancer cell line A549. Methods A549 human lung cancer cell lines were divided into 5 groups of 0, 0.5, 2.5, 5.0 and 8.0μmol / L sodium selenite. After cultured with different concentrations of sodium selenite for 24h and 48h, the cells were stained with immunocytochemistry Western blot was used to detect the effect of sodium selenite on the expressions of autophagy-related proteins Beclin1 and LC3. Results Human lung adenocarcinoma A549 cells were cultured with different concentrations of sodium selenite for 24 hours. After 48 hours, the number of cells decreased, the cells became swollen and became larger, the chromatin became agglutinated, the nuclei were pyknotic and broken. The results of immunocytochemistry and Western blotting showed that the average optical density (MOD) of Beclin1 and LC3 immunocytochemical staining in lung cancer A549 cells was significantly increased with the increase of sodium selenite concentration (P <0.05). Conclusion Sodium selenite can inhibit the growth of lung cancer A549 cells and induce autophagy in lung cancer A549 cells.
