目的了解氟致成骨细胞内质网应激状态时成骨细胞分化相关基因的表达情况。方法用原代培养的人成骨细胞建立染氟模型,流式细胞仪测定凋亡指数,并提取RNA后用PCR芯片分别检测未折叠蛋白反应和骨分化相关基因的表达情况。结果氟能够引起成骨细胞内质网应激,引起15个基因表达上调,1个基因表达下调,其中涉及内质网应激PERK、IRE1和ATF6三条信号途径。骨分化方面有32个基因表达上调,2个基因表达下调,涉及胶原、基质金属蛋白酶、整合素、骨形态发生蛋白、血管生长因子、肿瘤坏死因子等基因。结论氟引起成骨细胞内质网应激的同时,对骨转化基因的表达也产生影响。
Objective To investigate the expression of osteoblast differentiation-related genes in endoplasmic reticulum-induced osteoblasts during fluorosis. Methods The primary cultured human osteoblasts were used to establish the model of fluorosis. The apoptotic index was determined by flow cytometry. The mRNA expression of unfolded proteins and the gene expression of osteoclast differentiation were detected by PCR chip. Results Fluorine could induce endoplasmic reticulum stress in osteoblasts, which resulted in up-regulation of 15 genes and down-regulation of 1 gene, which involved three ERK, IRE1 and ATF6 signaling pathways. 32 genes were up-regulated in bone differentiation and 2 genes were down-regulated, involving genes of collagen, matrix metalloproteinase, integrins, bone morphogenetic protein, angiogenic growth factor and tumor necrosis factor. Conclusion Fluorine can induce the endoplasmic reticulum stress in osteoblasts and affect the gene expression of osteoclasts.
