目的:初步探讨对于直接或间接引起的肺内感染,炎症反应产生的白介素(IL)-22能否刺激肺泡Ⅱ型上皮细胞,并且诱导其生成、释放2型人β防御素(humanβ-defensin-2,HBD-2)发挥抗感染作用。方法:流式细胞术检测肺泡Ⅱ型上皮细胞细胞膜表面IL-22R表达水平。重组人IL-22(20 ng/ml)刺激肺泡Ⅱ型上皮细胞,RT-PCR检测不同时间(0、2、4、8、16、24 h)HBD-2表达情况。不同浓度的重组人IL-22(0、4、20、100、500 ng/ml)刺激肺泡Ⅱ型上皮细胞,RT-PCR检测HBD-2表达情况。结果:肺泡Ⅱ型上皮细胞膜表面IL-22R表达呈强阳性,阳性率达(99.90±0.13)%。比较6个时间点HBD-2表达差异,发现HBD-2表达具有时间依赖性。比较不同浓度IL-22处理下HBD-2表达差异,IL-22 100 ng/ml为最佳干预浓度,且HBD-2表达与IL-22浓度具有依赖关系。结论:IL-22能显著诱导肺泡上皮细胞生成并释放HBD-2,且具有时间和剂量依赖性。
OBJECTIVE: To investigate whether interleukin-22 (IL-22) produced by inflammatory response stimulates the generation of alveolar type II epithelial cells induced by direct or indirect pulmonary infection and releases human beta-defensin- 2, HBD-2) play an anti-infective effect. Methods: Flow cytometry was used to detect the expression of IL-22R on the membrane surface of type Ⅱ alveolar epithelial cells. The type Ⅱ alveolar epithelial cells were stimulated with recombinant human IL-22 (20 ng / ml), and the expression of HBD-2 was detected by RT-PCR at different time points (0, 2, 4, 8, 16, 24 h). Different concentrations of recombinant human IL-22 (0, 4, 20, 100, 500 ng / ml) stimulated type II alveolar epithelial cells and the expression of HBD-2 was detected by RT-PCR. Results: The positive expression rate of IL-22R on the surface of type Ⅱ alveolar epithelial cells was 99.90 ± 0.13%. HBD-2 expression differences were compared at 6 time points and it was found that HBD-2 expression was time-dependent. The difference of HBD-2 expression between different concentrations of IL-22 was compared. IL-22 100 ng / ml was the best intervention concentration, and the expression of HBD-2 was dependent on IL-22 concentration. Conclusion: IL-22 can significantly induce the generation of alveolar epithelial cells and release HBD-2 in a time and dose-dependent manner.
