目的:探讨青鹏膏治疗大鼠胶原性关节炎的作用及其可能机制。方法:制备Ⅱ型胶原诱导大鼠关节炎模型,分组给药,测定大鼠关节损伤症候指数及关节滑膜病理组织学变化,采用免疫组化方法检测关节滑膜IL-1β及TNF-α的表达;以活化巨噬细胞系THP-1为模型,检测青鹏膏提取物对THP-1细胞TNF-α、IL-1βmRNA表达的影响。结果:青鹏膏对胶原性关节炎动物模型的足肿胀有明显抑制作用,给药组在关节肿胀指数、关节腔IL-1β、TNF-α表达水平等方面比模型对照组有所降低;青鹏膏提取物可使THP-1经LPS刺激的升高的IL-1β和TNF-αmRNA表达水平下降。结论:青鹏膏具有改善大鼠胶原性关节炎状的作用,该作用可能与其降低关节滑膜IL-1β及TNF-α分泌及基因表达有关。
Objective: To investigate the effect of Qingpeng ointment on collagen-induced arthritis in rats and its possible mechanism. METHODS: Type II collagen-induced rat arthritis models were prepared and administered in groups. The symptom indexes of joint injury and the histopathological changes of synovial membranes were determined. The synovial membranes of IL-1β and TNF-α were detected by immunohistochemistry. The expression of TNF-α and IL-1β mRNA in THP-1 cells was detected by the macrophage cell line THP-1 as a model. RESULTS: Qingpeng Ointment had obvious inhibitory effect on paw swelling of collagenous arthritis animal model. In the administration group, the joint swelling index, IL-1β and TNF-α expression levels in the joint cavity were lower than those in the model control group; Pengpeng extract can increase the expression of elevated IL-1β and TNF-α mRNA in THP-1 stimulated by LPS. Conclusion: Qingpeng ointment can improve collagen-induced arthritis in rats, which may be related to the decrease of IL-1β and TNF-α secretion and gene expression in synovial membrane of rats.
