目的 观察染尘巨噬细胞上清液对人胚肺成纤维细胞热休克蛋白70(HSP 70)各亚型(HSP70-1、HSP70-2、HSP 70-hom)表达的影响.方法 采用大鼠巨噬细胞(AMs)和人胚肺成纤维细胞(HELF)构成体外模型,采用逆转录多聚酶链反应(RT-PCR)技术检测二氧化硅粉尘处理AMs 24 h的培养上清液刺激HELF细胞的HSP 70各亚型表达情况.同时用二氧化钛作为阴性对照,以不用粉尘处理的AM培养上清液作为正常对照.结果 HSP 70-1、HSP 70-2、HSP 70-hom基因表达与粉尘浓度的相关系数(R)依次为0.931(P<0.01)、0.923(P<0.01)、0.818(P<0.05),呈现明显的剂量-反应关系.结论 受染尘肺泡巨噬细胞上清液刺激的人胚肺成纤维细胞HSP 70各亚型在转录水平呈表达增加,且可能参与了矽尘致细胞氧化应激过程.“,”Objective To observe the expression of HSP70-1 ,HSP70-2,HSPT0-hom in human embryonic lung fibro-blasts (HELF) induced by supematants of alveolar macrophage (AMs) exposed to silica. Methods AMs and HELF of rats were formed in vitro model. The HSP70-1, HSP70-2, HSPT0-hom mRNA levels of HELF stimulated by supematants of AMs exposed to silica were detected by RT-PCR technology. Restdts Expression of HSP70-1, HSPT0-2, HSP70-hom could be induced by dust-exposed AMs and there was a clear dose-effect relationship between HSP70-1, HSPT0-2, HSP70-hom mRNA levels and silica concentration with the correlation coefficients of 0. 931 (P<0. 01), 0. 923 (P<0. 01), 0. 818 (P<0. 05), respectively. Conclusion HSP70-1, HSP70-2, HSP70-hom may play an antioxidant role in the process of oxi-dative stress induced by silica.
