目的进一步探索环境雌激素双酚A(BPA)影响大鼠睾丸支持细胞波形蛋白 (Vimentin)的机理。方法采用支持细胞体外原代培养、免疫组织化学以及mRNA原位杂交技术分析了BPA对支持细胞Vimentin及其基因转录、参与Vimentin降解的蛋白激酶C(PKC)表达的影响。结果BPA使支持细胞拉长 ;支持细胞在体外系统中良好地表达Vi mentin ,BPA使其基因转录抑制 ;PKC在支持细胞中不表达。结论环境雌激素BPA通过抑制基因转录而干扰Vi mentin合成 ,从而影响支持细胞骨架的正常形成 ,导致细胞形态改变 ;PKC不直接干扰支持细胞Vimentin代谢。
Objective To explore the mechanism of environment estrogen bisphenol A (BPA) affecting the vimentin in rat testicular supportive cells. Methods The effects of BPA on the expression of Vimentin and its gene transcription and the expression of protein kinase C (PKC) involved in the degradation of Vimentin were analyzed using primary cultured cells in vitro, immunohistochemistry and mRNA in situ hybridization. Results BPA prolonged the number of supporting cells. The supporting cells expressed Vi mentin well in vitro and BPA inhibited the gene transcription. PKC did not express in the supporting cells. CONCLUSION: Environmental estrogen BPA can interfere with the synthesis of Vi mentin by inhibiting gene transcription, thus affecting the normal formation of supporting cytoskeleton and leading to cell morphological changes. PKC does not directly interfere with the support of Vimentin metabolism.
