细胞的体积调节过程伴随着细胞骨架的调节作用.将细胞置于低渗溶液的环境中,细胞体积膨胀,在细胞体积调节过程中细胞表面力学性质可能发生变化.在实验中,我们利用低浓度PBS溶液处理Hela细胞,利用AFM测定细胞表面杨氏模量的变化,反映细胞表面弹性性质的变化.实验结果显示,在低渗环境中,细胞表面的杨氏模量随时间发生波动.我们利用纳米尺度的角锥形针尖和微米尺度的球形针尖分别测量低渗条件下细胞表面杨氏模量的变化,得到的数据结果表明,不同尺度下测得的杨氏模量变化趋势略有不同.“,”Cytoskeleton regulation is part of cell volume regulation process.In hypotonic external solutions,cells swell and properties of cell surface will change.In our experiment,we treated Hela cells with low concentration PBS and tested Young's modulus of target cells' surface by Atomic Force Microscope.The results showed that Young's modulus would change with time during the hypotonic process.Nanoscale pyramid tips and microscale spherical tips were used respectively to test Young's modulus change to study the changes under diverse scales.Results showed that the changing trends under different scales differed.
