组织化学研究,死后取材的时间是很重要的,死后时间过长,组织产生自溶,对组织中酶的观察产生一定影响。当然组织愈新鲜愈好,但限于条件一般要求取材不超过死后6小时。心肌组织在正常情况下溶酶体数量不多,自溶现象产生得比较慢,但在死后6小时取材对心肌组织化学酶的观察是否有影响则不清楚。为此我们设计了本实验,以观察大白鼠在死后2—12小时内心肌琥珀酸脱氢酶、酸性磷酸酶的变化。实验方法: 采用成年大白鼠20只,用拉脊椎方法处死,不放血,摹拟人死后情况,放置于室温18℃左右,除死后6小时杀死8只外,余在2小时、4小时、12小时各杀4只。每次处

Experimental methods: Twenty adult rats were sacrificed by pulling the spine, without bleeding. After the death was simulated, the animals were placed at room temperature of about 18 ℃.
