目的:观察MMP-2、MMP-9、VEGF和RECK基因在膀胱癌组织中的表达,探讨膀胱癌转移可能的基因机制。方法:采用RT-PCR法检测50例膀胱癌手术切除标本中MMP-2、MMP-9、VEGF和RECK基因的表达,并分析其临床意义。结果:MMP-2、MMP-9、VEGF和RECK在膀胱癌组织中的阳性表达率分别为80%、56%、78%和64%。膀胱癌组织MMP-2、MMP-9和VEGF三种基因的表达水平高于癌周组织(P<0.05),而RECK基因的表达水平低于癌周组织(P<0.05)。膀胱癌组织中MMP-2、MMP-9和VEGF基因的表达水平与膀胱癌组织的病理分级分期密切相关,而RECK基因的表达水平与膀胱癌的分级分期无明显相关。结论:膀胱癌组织存在多基因的表达异常,其发生、发展及转移与多基因联合作用有关。
OBJECTIVE: To investigate the expression of MMP-2, MMP-9, VEGF and RECK in bladder transitional cell carcinoma and to explore the possible gene mechanisms of bladder cancer metastasis. Methods: The expressions of MMP-2, MMP-9, VEGF and RECK in 50 specimens of bladder cancer were detected by RT-PCR and the clinical significance was analyzed. Results: The positive rates of MMP-2, MMP-9, VEGF and RECK in bladder cancer were 80%, 56%, 78% and 64% respectively. The expression of MMP-2, MMP-9 and VEGF in bladder cancer was higher than that in peri-cancerous tissues (P <0.05), while the expression level of RECK was lower than that in peritumoral tissues (P <0.05). The expression of MMP-2, MMP-9 and VEGF in bladder cancer was closely related to the pathological grading and staging of bladder cancer, while the expression of RECK was not related to the stage of bladder cancer. Conclusion: The expression of multiple genes in bladder cancer is abnormal. The occurrence, development and metastasis of bladder cancer are related to the combined effect of multiple genes.
