目的观察胃复宁煎剂对CAG模型大鼠胃黏膜异型增生及PCNA、EGF表达的调节作用以探讨其治疗机理。方法采用以MNNG诱导为主的综合造模法进行造模,为期24 W,造模结束后治疗12 W。分别予以生理盐水、胃复宁大小剂量组、维霉素组治疗。实验前后观察大鼠的体重变化并检测PCNA、EGF变化和组织病理学变化,进行比较分析。结果与空白组相比,模型组大鼠胃黏膜组织PCNA、EGF水平明显高于正常组(P<0.05),胃黏膜组织病变明显。治疗后各组大鼠胃黏膜组织PCNA、EGF水平明显降低,胃黏膜病理组织学变化明显改善。胃复宁大、小剂量组与模型组PCNA、EGF表达比较,有显著性差异(P<0.05);胃复宁大剂量组与胃复宁小剂量组PCNA、EGF表达比较,无显著性差异(P>0.05)。维霉素组与胃复宁大剂量治疗组PCNA、EGF表达比较,有明显差异(P<0.05)。结论胃复宁(尤其是大剂量组)通过对胃黏膜组织PCNA、EGF表达的干预作用,达到了对CAG大鼠胃黏膜细胞过度增殖的抑制作用,使部分萎缩或和增生的黏膜得到逆转,降低了胃癌发生的可能性。
Objective To observe the regulatory effect of Wei Fu Ning decoction on dysplasia of gastric mucosa and expression of PCNA and EGF in CAG model rats to explore its therapeutic mechanism. Methods A comprehensive modeling method based on MNNG induction was used for modeling, with a duration of 24 W and a treatment of 12 W after modeling. Respectively, saline, stomach Fu Ning size dose group, treatment of retinomycin. Before and after the experiment, we observed the changes of body weight, detected the changes of PCNA, EGF and histopathological changes, and compared them. Results Compared with the blank group, the levels of PCNA and EGF in gastric mucosa in model group were significantly higher than those in normal group (P <0.05), and the lesions in gastric mucosa were obvious. After treatment, the levels of PCNA and EGF in gastric mucosa of rats in each group were significantly decreased, and the histopathological changes of gastric mucosa were significantly improved. There was a significant difference in the expression of PCNA and EGF between the large and small doses of Weifu Ning group and the model group (P <0.05). There was no significant difference in the expression of PCNA and EGF between the high-dose and low-dose Weifenning groups (P> 0.05). The expression of PCNA and EGF were significantly different between the treatment group and the high-dose Weibu Ning group (P <0.05). Conclusion Gas Fu Ning (especially the high-dose group) can inhibit gastric mucosal hyperproliferation in CAG rats by intervening the expression of PCNA and EGF in gastric mucosa, reversing some of the atrophy or proliferating mucosa, Reduce the possibility of gastric cancer.
