小鼠Ⅳ型超敏反应模型建立的研究

本文应用旧结核菌素(OT)与福氏不完全佐剂混合后注射小鼠皮下,以诱导小鼠Ⅳ型超敏反应,并与牛血清白蛋白(BSA)注射组比较,以125I-UdR注射后检测局部血管通透性为指标,观察此两种抗原诱导Ⅳ型超敏反应的结果。结要表明,0.25u的OT两次注射后能明显导致Ⅳ型超敏反应,血清中仅检测到低水平抗体,而BSA组,第二次注射的局部未见明显血管通透性增加现象。病理切片观察亦显示相似结果。提示OT注射小鼠皮下建立第Ⅳ型超敏反应动物模型,具有方法简便、结果可靠、特异性高等特点。 In this paper, the mice were subcutaneously injected with old tuberculin (OT) and incomplete Freund’s adjuvant to induce the type IV hypersensitivity in mice. Compared with the BSA injection group, the mice were treated with 125I-UdR After injection of local vascular permeability as an indicator to observe the two antigens induced type Ⅳ hypersensitivity results. Results showed that 0.25u OT twice after injection can significantly lead to type Ⅳ hypersensitivity, serum only detected low levels of antibodies, and BSA group, the second injection of local no significant increase in vascular permeability . Pathological observations also showed similar results. Tip OT injected mice subcutaneously established type Ⅳ hypersensitivity animal model has the advantages of simple method, reliable results, high specificity.

从新生兔的四肢长骨中分离出破骨细胞,将其与大小约6mm×6mm、厚约10μm的牛骨片共同培养。24小时后,培养液中分别加入IL-2(50U/ml, 100U/ml);TNF-α(10-10M, 5×10-10M, 10-9M);PGE2(100ng/ml);并做空白对照。培养40小时、64小时及1周,利用相差显微镜计数吸收陷窝数,通过图像分析系统计算吸收陷窝表面积。结果表明:IL-2能促进破骨细胞
