目的鉴定红细胞(RBCs)中有无内皮型一氧化氮合酶(eNOS),探讨在4℃保存条件下,RBCs中的eNOS含量随保存时间变化的规律。方法采用免疫印迹(WB)和免疫荧光技术检测库存红细胞膜标本中有无eNOS;采用逆转录-PCR(RT-PCR)技术eNOS-mRNA库存红细胞中的检测。结果 RBCs中eNOS检测结果呈阳性。灰度分析结果表明:在RBCs保存35 d内,并不能引起eNOS含量的变化;而RBCs中并没有检测到eNOS-mRNA的存在。结论 RBCs含有eNOS;在库存血液35 d的保存期内,RBCs中的eNOS含量保持稳定。另外,在RBCs中未发现有eNOS-mRNA存在。
Objective To identify the presence or absence of endothelial nitric oxide synthase (eNOS) in erythrocytes (RBCs) and to investigate the regularity of eNOS content in RBCs with storage time at 4 ℃. Methods Immunocytochemistry (WB) and immunofluorescence were used to detect the presence or absence of eNOS in the stock samples of erythrocytes. The detection of eNOS-mRNA in the stock samples of erythrocytes was performed by reverse transcription-polymerase chain reaction (RT-PCR) Results The results of eNOS in RBCs were positive. The results of gray scale analysis showed that the changes of eNOS content did not result in the storage of RBCs within 35 days, while the presence of eNOS-mRNA was not detected in RBCs. Conclusions RBCs contain eNOS. The eNOS content in RBCs remained stable during the 35-day storage of blood in stock. In addition, no eNOS-mRNA was found in RBCs.
