一起食物中毒病因的实验室分析

目的查明引起食物中毒的致病菌,分析菌株间的亲缘关系,为明确食物中毒诊断提供依据。方法采集一起食物中毒蛋糕和病人样品,用实时荧光PCR快速筛检,按GB 4789.4—2010《食品微生物学检验沙门菌检验法》分离、鉴定致病菌,脉冲电场凝胶电泳(PFGE)对病原菌作同源性分析。结果从16份蛋糕样品中检出8株肠炎沙门菌,32份患者粪便样品中检出17株肠炎沙门菌,PCR核酸阳性与GB 4789.4—2010法分离到菌株完全一致;PFGE条带聚类分析显示患者与蛋糕中检出的肠炎沙门菌属同一基因型,具有高度同源性。结论本起食物中毒由肠炎沙门菌污染蛋糕所致;PCR法与GB 4789.4—2010法联合检测,有助于快速锁定食物中毒致病菌,从基因水平上证明食物病原的相关性。 Objective To identify the pathogenic bacteria that cause food poisoning, analyze the genetic relationship among the strains and provide evidence for the diagnosis of food poisoning. Methods Samples of food poisoning cake and patient were collected and quickly screened by real-time fluorescence PCR. The pathogens were isolated and identified according to GB 4789.4-2010 “Food Microbiological Test Salmonella Test”, and the effect of PFGE on pathogenic bacteria Homology analysis. Results Eight strains of Salmonella enteritidis were detected in 16 samples of cake, and 17 strains of Salmonella enteritidis were detected in 32 samples of stool samples. The PCR nucleic acid positive was identical to that of strain GB 4789.4-2010. The PFGE bands clustering analysis Patients and the cake showed the same genotype of Salmonella enteritis, with a high degree of homology. Conclusion The food poisoning caused by Salmonella enteritidis contamination of the cake caused; PCR method and GB 4789.4-2010 joint detection, help to quickly lock food poisoning pathogens, from the genetic level to prove the relevance of food pathogens.
