目的:评估灯盏花中灯盏花乙素超声提取法的有效性,优化超声提取的条件。方法:在单因素试验的基础上,选取超声波作用时间、液料比和乙醇浓度为影响因子,采用星点设计(CCD)进行3因素3水平的实验设计,以HPLC测定灯盏花乙素得率作为响应值,进行响应面分析(RSM)。结果:最佳提取条件为:2.0 g灯盏花原料,超声提取时间24.5 min,乙醇浓度74.7%,液料比19.8 mL/g,二次提取得率预测值为0.641%,验证值为0.632%(n=3),优于单因素优化结果,与预测值的相对误差为-1.14%。结论:超声提取是种简单、有效、低成本的方法,可改善灯盏花中灯盏花乙素的提取过程。星点设计-效应面法优选预测值与实际值吻合度高。
OBJECTIVE: To evaluate the effectiveness of sonobuy supersonic extraction in erigeron and optimize the conditions of sonication extraction. Methods: On the basis of single factor experiment, ultrasonic effect time, liquid material ratio and ethanol concentration were selected as the influencing factors. The experiment design of 3 factors and 3 levels was carried out by using the star point design (CCD). The yield of scutellarin As a response, response surface analysis (RSM) was performed. Results: The optimum extraction conditions were as follows: 2.0 g of Erigeron breviscapus raw material, ultrasonic extraction time 24.5 min, ethanol concentration 74.7%, liquid to liquid ratio 19.8 mL / g, the second extraction yield was 0.641%, the validation value was 0.632% n = 3), which is better than the result of single factor optimization. The relative error with the predicted value is -1.14%. Conclusion: Ultrasonic extraction is a simple, effective and low cost method to improve the extraction process of scutellarin in Erigeron.
